For transport you need, like cradle storing, the full weight on the keel, 
but the screw jacks tighter, but not deflecting the hull

When on a cradle I have the screw jacks snug enough to prevent toppling but 
not actually deflecting the hull and can, with some effort, twist the pads 
themselves by hand.

You should then secure the boat with robust tie down straps over the top or 
to the toe rail down to the trailer ensuring they do not mar the hull before 
trailering so the boat can't bounce.

I can't comment on going through the aperture because I have never done that 
and am not sure about the glass thickness layup there

Yes that is the horn timber above and slightly behind the rudder curving 
aft.
